{"created":"2023-06-23T12:19:44.024794+00:00","id":10823,"links":{},"metadata":{"_buckets":{"deposit":"cbe24828-5ce7-4557-ba28-7a1cfa689852"},"_deposit":{"created_by":21,"id":"10823","owners":[21],"pid":{"revision_id":0,"type":"depid","value":"10823"},"status":"published"},"_oai":{"id":"oai:asahi-u.repo.nii.ac.jp:00010823","sets":["46:348:431"]},"author_link":["3934","4131","3930","3931","4171","4172","1776"],"item_10002_biblio_info_77":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2019-02","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"3","bibliographicPageEnd":"148","bibliographicPageStart":"141","bibliographicVolumeNumber":"45","bibliographic_titles":[{"bibliographic_title":"岐阜歯科学会雑誌"},{"bibliographic_title":"The Journal of Gifu Dental Society","bibliographic_titleLang":"en"}]}]},"item_10002_description_80":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"我々は以前、歯周病関連細菌の有する外膜タンパク質にO-結合型のN-アセチルグルコサミン（GlcNAc）やシアル酸が付加されていることを明らかにしてきた。その一方で歯周病関連細菌の糖タンパク質の役割については未だ不明な点が多い。本研究では、哺乳類細胞において糖タンパク質へのGlcNAc付加を仲介するGlcNAc転移酵素の阻害剤、ならびにシアル酸付加を仲介するシアル酸転移酵素の阻害剤を用い、これらが歯周病関連細菌Porphyromonas gingivalisの増殖、血清抵抗性ならびに糖タンパク質合成に対してどのような影響を及ぼすのか調べたので報告する。菌株はP. gingivalis ATCC 33277を用いた。GlcNAc転移酵素阻害剤としてBADGPおよびOSMI-1、シアル酸転移酵素阻害剤として3-Fax-Peracetyl Neu5AcおよびSoyasaponin Iを用いた。各阻害剤のP. gingivalisの増殖への影響は、阻害剤を添加した細菌用培地で菌株を培養後にATP産生量を測定して調べた。同様にsTSB培地に40%ヒト血清を加えた場合のP. gingivalisの増殖への各阻害剤の影響も調べた。また各阻害剤の存在下で増殖させたP. gingivalisの菌体は破砕してSDS-PAGEで展開後、糖タンパク質をPro-Q Emerald 300染色により検出し、阻害剤の影響を解析した。細菌用培地でP. gingivalisを培養した場合、OSMI-1とSoyasaponin IでP. gingivalisの増殖がわずかに促進された。一方、血清を添加した培地で培養した場合には、BADGPのみ増殖を抑制したが、他の阻害剤では増殖が大きく促進された。これらの結果に反して、阻害剤によりP. gingivalisの糖タンパク質の発現パターンに明確な変化は認められなかった。以上の結果より、糖鎖修飾関連酵素阻害剤はP. gingivalisの糖タンパク質産生には影響しないが、何らかの非特異的効果によって増殖に影響すると考えられた。","subitem_description_type":"Abstract"}]},"item_10002_description_81":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"We have previously reported that periodontopathic bacteria possess outer membrane proteins that are linked to O-linked N-acetyl glucosamine (O-GlcNAc) and sialic acid. However, specific roles of these glycoproteins in the pathogenicity of periodontopathic bacteria have been remained largely unclear. In this study, we examined the effect of mammalian glycosyltransferase inhibitors̶specifically, GlcNAc transferase inhibitors and sialyltransferase inhibitors, on the growth, serum resistance, and glycoprotein synthesis of Porphyromonas gingivalis. The strain of P. gingivalis ATCC 33277 was used in this study. BADGP and OSMI-1 were used as the GlcNAc transferase inhibitors, and 3-Fax-Peracetyl Neu5Ac and Soyasaponin I were used as the sialyltransferase inhibitors. Bacteria were cultured in sTSB or 40% inactivated bovine serum-containing sTSB supplemented with one of the inhibitors. Bacterial growth was monitored by measuring ATP production. Whole bacterial lysates were separated by SDS-PAGE, and glycoproteins were stained using Pro-Q emerald 300. In the bacterial medium lacking serum, OSMI-1 and Soyasaponin I were found to slightly enhance the growth of P. gingivalis. Interestingly, BADGP was found to have a suppressive effect on bacterial growth in serum-containing bacterial medium, whereas other inhibitors in the same serum-conFaxtaining medium greatly enhanced bacterial growth. Our studies did not detect any alteration in glycoprotein synthesis in the presence of the inhibitors. These findings indicate that mammalian glycosyltransferase inhibitors do not aff ect glycoprotein synthesis of P. gingivalis, but they exert significant regulatory effects on the bacterial growth possibly through non-specific
