{"created":"2023-06-23T12:21:52.388345+00:00","id":13598,"links":{},"metadata":{"_buckets":{"deposit":"7d6b0c1d-97a0-4e83-a09b-be9eb7786cca"},"_deposit":{"created_by":21,"id":"13598","owners":[21],"pid":{"revision_id":0,"type":"depid","value":"13598"},"status":"published"},"_oai":{"id":"oai:asahi-u.repo.nii.ac.jp:00013598","sets":["46:190:414"]},"author_link":["4349","4380","3971","4382","3788","4353","4377","549","4385"],"item_10002_biblio_info_77":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2001-06-20","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"1","bibliographicPageEnd":"101","bibliographicPageStart":"90","bibliographicVolumeNumber":"28","bibliographic_titles":[{"bibliographic_title":"岐阜歯科学会雑誌"},{"bibliographic_title":"The journal of Gifu Dental Society","bibliographic_titleLang":"en"}]}]},"item_10002_description_80":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"炭酸イオンを含有したアパタイト焼結体(CAp)の骨補填材としての有用性を評価するために,ラットから採取した骨芽細胞様細胞をCAp基盤上で培養し,骨基質形成に及ぼす基盤の影響を検討した.比較基盤として,水酸化アパタイト(HAp),β-リン酸三カルシウム(β-TCP),マグネシウムを添加し高温安定化したリン酸三カルシウム(Mg-β-TCP),HApとMg-β-TCPを混合し(HAp-Mg, β-TCP)作製した基盤,および培養用スチロールプレートを用いた.形状および表面性状をほぼ同一とした各基盤上に4-5週齢Wistar系ラットから採取した培養骨芽細胞様細胞を播種し,8日及び14日間培養し,アルカリホスファターゼ活性(ALP活性),DNA量及び石灰化物に起因するカルシウム及びリン酸濃度を測定した.また,各基盤表面のSEM観察および剥離組織のEPMA観察ならびにX線回折も行い,基盤表面に沈着した石灰化相の同定も行った.培養8日目では,β-TCPはCAp, HAp, Mg-β-TCP, HAp-Mg, β-TCPよりALP活性が低い傾向を示すものの,DNA量に関しては各リン酸カルシウム系基盤間で大きな差異は認められなかった.しかし,スチロール基盤に比べ,各リン酸カルシウム系基盤はALP活性およびDNA量で大きな値を示し,細胞の分化・増殖がリン酸カルシウム系基盤では助長されることが示唆できた.また,カルシウムおよびリン酸濃度の基盤依存性はALP活性およびDNA量の基盤依存性と類似し,リン酸カルシウム系基盤上では石灰化相の形成も助長されることが示された.培養14日目になると,各基盤についてALP活性,DNA量,カルシウムおよびリン酸濃度は上昇するが,スチロール基盤では,上昇程度が相対的に高く,リン酸カルシウム系基盤上では細胞がコンフルエントな状態になっていることが推察された.しかしながら,β-TCPではALP活性およびDNA量が他のリン酸カルシウム系基盤に比べ有意に低く,また,Mg-β-TCPではカルシウム及びリン酸濃度が低い値を示す傾向にあった.これに反し,CApはHApおよびHAp-Mg, β-TCPと遜色なく,細胞の分化・増殖および石灰化相を含む基質形成が良好で,骨補填材として有用な材料となることが確認できた.","subitem_description_type":"Abstract"}]},"item_10002_description_81":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"In rat-marrow-cell cultures, sintered carbonate apative (CAp) as a bone substitute was evaluated, investigating the ability of marrow cells to proliferate and to produce a mineralized matrix on this substrate. In these cell cultures, calcium phosphate bioceramics such as hydroxyapatite (HAp), β-tricalcium phosphate (β-TCP), magnesium containing β-tricalcium phosphate (Mg, β-TCP), a mixture of hydroxyapatite and magnesium containing β-tricalcium phosphate (HAp-Mg, β-TCP), and polystyrene, which is a material used as plating dishes, were compared with CAp. When compared at 8 days of culture, the number of cells, which could be estimated by DNA content, was significantly greater with calcium phosphate substrates, irrespective of the difference in compositions, than with polystyrene substrates. The dependence of ALPase activity on substrates was essentially the same with that of DNA content, suggesting that bone marrow cells proliferate and differentiate similarly on any type of calcium phosphate substrates. In addition, no significant differences were observed in the morphology of cells on calcium phosphate substrates, on which they appeared to be spread and to contact with their borders, in contrast to the spherical appearance with the control substrates. With the exception of the polystyrene substrate, fibral extracellular matrix was evident on calcium phosphate substrates. ALPase activity as well as DNA content doubled approximately at 14 days of culture for all substrates made of calcium phosphates except β-TCP, with which they were about 20% lower than the averaged values. With polystyrene substrates, however, both the ALPase activity and DNA content increased to an extent that was comparable with HAp, CAp, β-TCP. and HAp-Mg, β-TCP substrates. in addition, calcium and phosphate contents had markedly increased by day 14 for all substrates including polystyrene. The results showed that CAp is as effective as HAp and HAp-Mg, β-TCP as a material to support cell
