{"created":"2023-06-23T12:22:11.310992+00:00","id":13997,"links":{},"metadata":{"_buckets":{"deposit":"2d1c0fc4-6f9b-4c7e-a3e9-d44e8ddc38c0"},"_deposit":{"created_by":21,"id":"13997","owners":[21],"pid":{"revision_id":0,"type":"depid","value":"13997"},"status":"published"},"_oai":{"id":"oai:asahi-u.repo.nii.ac.jp:00013997","sets":["46:190:423"]},"author_link":["4956","5529","4955","16817"],"item_10002_biblio_info_77":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2004-11-20","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"特集","bibliographicPageEnd":"163","bibliographicPageStart":"156","bibliographicVolumeNumber":"30","bibliographic_titles":[{"bibliographic_title":"岐阜歯科学会雑誌"},{"bibliographic_title":"The journal of Gifu Dental Society","bibliographic_titleLang":"en"}]}]},"item_10002_description_80":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"らせん状細菌である口腔トレポネーマは,歯周病患者の歯肉縁下プラークより高頻度に分離されるが,その病態形成における役割についてはほとんど明らかにされていない.本稿において,これら口腔トレポネーマの菌体表層成分の構造と機能についてその一端を明らかにした.すなわち,歯周病患者における歯肉縁下プラーク中の口腔スピロヘータTreponema denticola, Treponema vincentiiおよびTreponema mediumの分布について,各菌特異的な16S rRNAプライマーを用いたPCR法により検討した.その結果,これら口腔トレポネーマの検出率は, T. denticola, 68.0%; T. vincentii, 36.0%; T. medium, 48.0%であり,健常者と比較して歯周病患者では検出率が高くなる傾向がみられた.なかでも,T. denticolaおよびT. mediumは,歯周ポケットが深くなるとともに検出率も上昇した.ついでこれら口腔トレポネーマのなかで,T. medium ATCC 700293^T菌体より複合糖質画分(Tm-Gp)を抽出・精製し,その化学構造と免疫生物学的作用について検討した.Tm-Gpは複合糖質(Tm-GC)と多糖部分(Tm-PS)から構成され,Tm-GCはTm-PSにホスファチジルグリセロールが結合したものであった.また,Tm-PSは4糖2アミノ酸を繰り返し構造とする多糖で,その化学構造は[→4)β-D-GlcpNAc3NAcA (1→4)β-D-ManpNAc3NAOrn (1→3)β-D-GlcpNAc (1→3)α-D-Fucp4NAsp (1→]であり,グラム陰性細菌由来のリポ多糖(LPS)やグラム陽性菌由来のリポタイコ酸とは異なる構造であった.Tm-Gpは,LPS様の内毒素活性はみられなかったが,LPSによるヒト単球細胞株Mono Mac 6の活性化を濃度依存的に抑制した.また,Tm-Gpは,LPSやその活性中心であるリピドAによるマウスToll様受容体(TLR) 4/MD-2強制発現マウスB細胞株Ba/F3のNF-_κB活性を抑制したが,Taxolによる同活性の抑制はみられなかった.さらにTm-Gpの抑制作用は細胞培養系における血清無添加の条件下ではみられず,血清中に存在するCD14やLPS結合タンパク(LBP)の添加により示された.また,Tm-Gpは,固相化CD14あるいはLBPへのLPS結合を阻害した.これら抑制作用は,T. medium複合糖質中のホスファチジルグリセロールによって担われていることを明らかにした.以上の結果から,歯周病の病態が悪化するとともに増加する口腔トレポネーマ,なかでもT. medium由来複合糖質は,宿主の自然免疫応答における抑制因子として作用し,歯周病患者の病巣局所における慢性化に関与することが考えられる.","subitem_description_type":"Abstract"}]},"item_10002_description_81":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"Spirochetes are well-known as causative agents of various chronic infectious diseases. In the present study, we developed a highly sensitive and specific method for detecting and quantifying the cultivable oral treponemes, such as Treponema denticola, Treponema vincentii and Treponema medium in subgingival plaque samples from patients with periodontal diseases, using PCR assays with specific primers for each 16S rRNA sequence. The frequencies of occurrence for the three oral treponemes in the subgingival plaque samples were 68.0%, 36.0%, and 48.0%, respectively. In addition, the numbers of T. medium and T. denticola cells detected in the plaque samples were greater in samples taken from deep periodontal pockets. These findings suggest that various oral treponemes are associated with the formation of periodontal disease stages. Next, we separated a glycoconjugate preparation from T. medium ATCC 700293^T (Tm-Gp), and then determined its chemical structure and immunobiological activities. Tm-Gp was found to consist of a glycoconjugate (Tm-GC) and a polysaccharide (Tm-PS), and Tm-GC was the structure that Tm-PS combined with a phosphatidylglycerol (PG). The chemical structure of Tm-PS was shown to be a tetrasaccharide backbone with two amino acids: [→4) β-D-Glc pNAc3NAcA (1→4) β-D-Man pNAc3 NAOrn (1→3) β-D-Glc pNAc (1→3) α-D-Fuc p4NAsp (1→], where GlcNAc3NAcA is 2, 3-diacetamido-2, 3-dideoxyglucuronic acid, ManNAc3NAOrn is N^d (2-acetamido-3-amino-2, 3-dideoxymannuronyl) ornithine, and Fuc4NAsp is 4-(α-aspartyl) amino-4, 6-dideoxygalactose. Tm-Gp diminished the activation of the human monocytic cell line Mono Mac 6 induced by lipopolysaccharide (LPS), but not that induced by TNF-α. NF-κB activation in Ba/F3 cells expressing mouse Toll-like receptor (TLR) 4 and MD-2, stimulated with LPS and its active center lipid A, but not with Taxol, was definitely inhibited by Tm-Gp, and was dependent on CD14/LPS-binding protein (LBP). Tm-Gp blocked the binding of LPS to immobilized CD14 and LBP, and also inhibited nitric oxide production in a mouse macrophage cell line, J774-A1, stimulated with various bacterial components in the presence of FBS. Further-more, the inhibitory effects seemed to be attributable to the lipophilic part (PG) of the glycoconjugate from Tm-GC. These results indicate that T. medium, which contains a glycoconjugate possessing an inhibitory effect toward TLR-mediated cell activation, may act as a modulator of innate immune
