{"created":"2023-06-23T12:22:14.898360+00:00","id":14068,"links":{},"metadata":{"_buckets":{"deposit":"c77290b0-9cf6-4d14-86f7-3cdbba358e35"},"_deposit":{"created_by":21,"id":"14068","owners":[21],"pid":{"revision_id":0,"type":"depid","value":"14068"},"status":"published"},"_oai":{"id":"oai:asahi-u.repo.nii.ac.jp:00014068","sets":["46:190:428"]},"author_link":["6262","8724","6258","8727","345","8728","535"],"item_10002_biblio_info_77":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2006-06-25","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"1","bibliographicPageEnd":"46","bibliographicPageStart":"39","bibliographicVolumeNumber":"33","bibliographic_titles":[{"bibliographic_title":"岐阜歯科学会雑誌"},{"bibliographic_title":"The Journal of Gifu Dental Society","bibliographic_titleLang":"en"}]}]},"item_10002_description_80":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"エナメル質形成におけるGタンパクおよびそれに関わるレセプター、細胞内シグナル伝達の局在を明らかにするため、ラット臼歯を用いてGsα、Giα、L型Caイオンチャンネル、Goα、副甲状腺ホルモンレセプター(PTH-R)、Gqα、血管上皮成長因子(VEGF-R)、さらにホスホリパーゼC(PLCβ)の局在性を免疫組織化学的に検討した。成熟期では、成熟期エナメル芽細胞、中間層細胞さらに乳頭層細胞におけるGsαの発現は、それらの細胞内においてGiαによる抑制を受けずに、細胞機能に関わっていることが推測された。GsαとカップリングするPTH-Rは、分化期から分泌期エナメル芽細胞の近心端部、さらに成熟期エナメル芽細胞の近心端部のみにやや強い発現性を認め、Gsαとの共在性が認められた。PTH-Rのエナメル芽細胞における発現を認めたことは、従来の機能的な実験手法による結果を加味して、GsαとカップリングするPTH-Rは分化期、分泌期、成熟期エナメル芽細胞の機能発現に関わることが推察される。Gsαとの共在性を認めた分化期、分泌期初期のエナメル芽細胞、さらに分泌期後期エナメル芽細胞にはL型Caイオンチャンネルが発現し、細胞内へのCa^<2+>導入に関わることが示唆される。Gqαを介するレセプターや細胞膜情報伝達系が分化期、分泌期さらに成熟期のエナメル芽細胞と隣接する中間層細胞、乳頭層細胞において、細胞内のCa^<2+>動員を行い、各発育時期での機能発現に関わっていると考えられる。VEGF-Rは、Gqαと同様に内エナメル上皮ではほとんど染色性を認めないが、分化期から分泌期エナメル芽細胞の近遠心端部に強い染色性を認め、Gqαとほぼ同様な染色性を認めた。PLCβは内エナメル上皮、分化期エナメル芽細胞、分泌期エナメル芽細胞の近心端部および隣接する中間層細胞すべてに同程度の染色性を認めた。特に分化期および分泌期エナメル芽細胞においてGqαとPLCβと同様の染色性が認められた。以上より、Gタンパクとそれらに関わるCaイオンチャンネルおよびレセプターはエナメル質形成におけるエナメル芽細胞の各発育時期においてそれぞれの役割や機能に関わることが示唆された。","subitem_description_type":"Abstract"}]},"item_10002_description_81":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"The purpose of this study was to clarify the localization of the receptor and signal-transduction pathway linkages related to amelogenesis in developing rat molars, Gsα, Giα, L-type calcium channel, Goα, parathormone receptor (PTH-R), Gqα, vascular endothelial growth factor receptor (VEGF-R) and phospholipase C (PLCẞ) were examined immunohistochemically. In the maturation stage of ameloblasts, Gsα localization showed that the cells of stratum intermedium and papillary layer were not associated with Giα. PTH-R was stained slightly stronger at only the mesial end of presecretory ameloblasts from the maturation stage to the differentiation stage, and localization was essentially the same with Gsα. The appearance of PTH-R, with coexistiat with Gsα, suggested that the function of the manifestration of ameloblasts in the differention stage, secretory stage and also the maturation stage was closely associated with PTH-R. In the differentiation stage and secretory stage of ameloblasts coupled with Gsα, L-type calcium channels appeared and contributed to the influx of Ca^<2+> inside the cells. A receptor through Gqα and the cell membrane communication system have been reported to be involved in the function manifestration of ameloblasts by mobilizing intercellular Ca^<2+> at every stage and adjacent to the cells of the stratum intermedium, papillary layer cells. The VEGF receptor as well as Gqα was not present in the inner dental epithelium, but it showed strong stainability at the mesial and distal ends of ameloblasts in the secretory stage. PLCẞ appeard in the secretory stage, inner dental epithelium, differentiation stage, the mesial end of ameloblasts and adjacent to the cells of the stratum intermedium. The stainability of Gqα was similar to that of PLCẞ, in particular in ameloblasts in the differentiation stage and secretory stage. These results suggest that G proteins, PTH-R and L-type calcium channel associated with the protein, play important roles in each developmental stage of ameloblasts.
