{"created":"2023-06-23T12:18:33.557763+00:00","id":9234,"links":{},"metadata":{"_buckets":{"deposit":"e8c8dcc5-6b40-4ee4-992b-48ff88eb7865"},"_deposit":{"created_by":21,"id":"9234","owners":[21],"pid":{"revision_id":0,"type":"depid","value":"9234"},"status":"published"},"_oai":{"id":"oai:asahi-u.repo.nii.ac.jp:00009234","sets":["46:348:376"]},"author_link":["3934","3931"],"item_10002_biblio_info_77":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2018-01","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"2","bibliographicPageEnd":"158","bibliographicPageStart":"145","bibliographicVolumeNumber":"44","bibliographic_titles":[{"bibliographic_title":"岐阜歯科学会雑誌"},{"bibliographic_title":"The Journal of Gifu Dental Society","bibliographic_titleLang":"en"}]}]},"item_10002_description_80":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"病原体が組織に感染を起こすと、自然免疫系は病原体の存在をパターン認識受容体(PRR)を使って感知し、炎症反応を誘導して病原体の排除や組織の修復を進めていく。一方、病原体の存在が無い、いわゆる「非感染性炎症」の場合でもPRRによる自然免疫系の認識機構が重要な役割を果たしている。近年、自然免疫系による認識を受けて炎症反応を惹起する内因性分子が次々と同定されており、「ダメージ関連分子パターン(DAMP)」と呼ばれている。その機能や役割に関して、免疫学的、生理学的、あるいは病理学的意義について注目が集まっている。DAMPとして働く分子の多くは通常、組織中や細胞内に格納されており、PRRによる認識から物理的に隔離されている。DAMPの放出・遊離はストレスや損傷による組織障害、あるいは細胞破壊や細胞死に伴い、組織中や細胞内の空間から二次的に生じてくる。DAMPは生体の危機を知らせる「危険シグナル」として生理学的に重要な役割をもつだけでなく、過剰なDAMPの生成は炎症を慢性化させ、あるいは生命を脅かすような炎症反応を引き起こすこともあり、病理学的にも重要な意味を持っている。本総説ではDAMPの由来と生成機序、PRRによる認識機構、誘導されるシグナル伝達経路について、著者が最近報告した歯周病に関連するDAMPであるγ-GTPの知見も含めて概説しながら非感染性炎症との関連性について考えていきたい。\n When host tissues suffer infections caused by pathogens, the innate immune system senses their presence through pattern recognition receptors (PRRs), and subsequently, initiates inflammatory responses whereby pathogen exclusion and tissue repair can be promoted. Additionally, the recognition mechanism of the innate immune system involving PRRs also plays an important role in “sterile infl ammation” that occurs in the absence of pathogens. In recent years, various endogenous molecules that induce inflammatory responses following recognition by the innate immune system have been identified, and are called damage-associated molecular patterns (DAMPs). Considerable attention has been focused on their functions and roles in inflammatory responses to investigate their immunological, physiological, or pathological significance. Most of the molecules acting as DAMPs are normally stored within tissues or cells, and therefore, they are physically isolated from PRR recognition. DAMP release from tissues and intracellular spaces occurs when tissues and cells are damaged by tissue stressors/injury and cell destruction/death, respectively. Thus, DAMPs have physiological functions as “danger signals” to inform the crisis of organisms. Moreover, excessive DAMP generation exerts pathological effects, and causes chronic inflammation or severe life-threatening infl ammatory reactions.
