値 Although diabetic cardiomyopathy, which develops in the absence of epicardial coronary artery disease, causes heart failure in diabetes, the pathogenesis is still unclear. Resveratrol has been reported to show a strong autophagy accelerating effect and thereby mitigate postinfarction heart failure. Here we investigated the possible therapeutic effect of resveratrol on diabetic cardiomyopathy in a model of type II diabetes. We used C57BL/KsJ-leprdb/leprdb (db/db) diabetes and C57BL/KsJ-leprdb/lepr- (db/-) non-diabetes control mice. Ten weeks old mice of each group were assigned into followings and treated for 2weeks; saline, resveratrol (50 mg/kg/day), chloroquine (autophagy inhibitor; 10 mg/kg/day). Non-diabetes db/- mice were assigned as well. In diabetes mice, although systolic cardiac function and geometry were similar to non-diabetes mice, diastolic dysfunction was apparent and severer compared with non-diabetes mice. In diabetes hearts, expression of LC3-II, p62 and phosphorylated mTOR was increased, while cathepsin D was decreased significantly. Ultrastructurally there were only few lysosomes in contrast with abundant autophagic vacuoles. These findings imply that autophagic process was suppressed at the final digestion step in diabetes hearts. Resveratrol or chloroquine did not influence the cardiac performance in non-diabetes mice. In diabetes mice, however, resveratrol mitigated diastolic dysfunction accompanying significantly augmented autophagic findings while chloroquine oppositely brought about a detrimental result. In conclusion, resveratrol may be a beneficial against diabetic cardiomyopathy, probably through its autophagy accelerating effect.
