| 内容記述 | 妊娠中のストレスは母体だけでなく胎児にも様々な影響を及ぼし、妊娠期のストレスによって出生後の子供の疾病や肥満発症のリスクが高まることも知られている。咀嚼運動には食物摂取の最初の段階である食物を細かく砕くという働き以外にも様々な効果が知られており、ストレス緩和作用もその一つである。本研究は妊娠中のストレス下での咀嚼運動が出生後の仔の肥満にどのような影響を及ぼすかを明らかにすることを目的とした。妊娠マウスをストレス群、ストレス／咀嚼運動群、対照群の3群に分けた。ストレス群には拘束ストレスを負荷し、ストレス／咀嚼運動群には拘束ストレスと同時に咀嚼運動を行わせた。ストレス負荷と咀嚼運動は妊娠12日目から出産まで行った。3群の母マウスから生まれた雄性仔マウスをそれぞれS群、S/C群、C群とし、離乳後、仔マウスには高脂肪飼料を与え16週齢まで飼育した。離乳後の仔マウスは、1週間ごとに体重と摂食量を測定し、脂肪量と血中アディポネクチン濃度については、16週齢のマウスについて測定した。16週齢のマウスでは、S群の脂肪量はC群と比較して有意に増加し、S/C群の脂肪量はS群よりやや減少した。また、血中アディポネクチン濃度はC群と比較してS群で有意に減少し、S/C群の血中濃度はS群より増加する傾向がみられた。これらの結果より、妊娠中のストレス下での咀嚼運動は、出生後の仔マウスの肥満を抑制する効果を示す可能性が示唆された。 Previous studies indicated that stress during pregnancy had negative consequences, such as learning deficits, diabetes and obesity, for both the mother and the off spring. Chewing is the initial action for food intake and is also an effective stress-coping behavior. We investigated effects of the prenatal stress and chewing on obesity risk in adult mouse off spring. Pregnant mice were randomly divided into control, stress, and stress/chewing groups. Stress was induced by placing mouse in a ventilated restraint tube, and was initiated on the day 12 of pregnancy and continued until delivery. Mice in the stress/chewing group were given a wooden stick to chew during restraint. Weaned male pups were fed a high-fat diet until 16 weeks of age. The body weights and food intakes were measured weekly. At the end of the experiment, the mouse fat weights and serum adiponectin levels were assessed. Prenatal stress led to increased fat weights and decreased serum adiponectin levels in adult off spring. Maternal chewing during stress attenuated stress-induced the elevated fat weights and reduced adiponectin levels. These findings suggest that maternal chewing during prenatal stress in dams could prevent obesity risk in their off spring. |
